Hello I have a question. I have some experience in .NET 4.0 but Im being hired for a job that requieres 2.0, is there much of a difference, will i have trouble adapting?
Also I have experience with MSSQL 2008 and they require MSSQL 2005, is there a big difference between them?

SQL Server shouldn't be a big issue. You'll get irritated with a few items missing in 2005.
If you are using the advanced new features of 4.0 in your design patterns, then yes, 2.0 will be a big issue. Generics came out in 2.0 so at least you have that. WCF not being around for 2.0 is a big deal too.

It is not a big deal, if any body have latest version exp. than they will easily catchup older one, only the matter is if you trying to implement new feture in 2.0 that are not supported than you will little difficult to find the way to complete such type of work in older version.
